Abstract

Background:
Human sperm chromatin and DNA integrity are susceptible to a variety of endogenous
and exogenous factors with different mechanisms throughout sperm production and transport, including DNA compaction and nucleoproteins. In order to initiate and maintain a viable pregnancy, the integrity of the paternal genome in both normal and assisted conception is of paramount importance
Aim:
Evaluation of the relationship between Sperm DNA integrity and Sperm chromatin
condensation and their relation to sperm parameters
Material and Mothed:
The current study includes semen samples100 Men attend the fertility clinic at the Higher Institute of Infertility Diagnostics and Assisted Reproductive Techniques / Al-Nahrain
University and Babel Teaching Hospital for Women and Children of normozoospermic and
patients with mild to moderate abnormalities according to WHO (1999). the sample is
divided into three categories, first the semen test according to WHO standards (1999), and
secondly the DNA integrity test using DNA fragmentation test and the third is the test
Condensation of chromatin using aniline blue dye.
Result:
In Correlation of DFI and sperm chromatin to seminal fluid characteristics, it was
significantly negatively correlated to sperm count/ ejaculate (r = - 0.223, p = 0.026) and DFI,
sperm chromatin was positively correlated to round cell count in a highly significant manner
(r = 0.39, < 0.001). The correlation of DFI with sperm chromatin in all infertile men was
positive and highly significant (r = 0.958; p < 0.001).
Conclusion:
1. In all enrolled men, DNA fragmentation was significantly negatively correlated to sperm
count/ ejaculate, positively correlated to round cell count and not significantly correlated to
other parameters.
2. In all enrolled men, sperm chromatin was significantly positively correlated to duration of
abstinence and positively correlated to round cell count.
3.The DNA fragmentation and sperm chromatin were being positively correlated. This could
make a suggestion that one test could be used instead of the other one.
