Light and Electron Microscopic Analysis of Rat`s Cerebellum Treated with Monosodium Glutamate

Abstract

Background: The cerebellum plays an essential role in preserving balance and movement coordination. Monosodium glutamate (MSG) is a widely used foods additive and was described to have many harmful effects.


Objective: Our study aimed to assess the potential toxic effect of MSG on the histological structure of the cerebellar cortex of albino rats.


Materials & Methods: Twenty rats were divided equally into two groups: the control group and the MSG treated group that were injected intraperitoneal with 3.5mg/g/day of MSG for 10 days, then cerebellar sections were obtained and prepared for light and electron microscopy.


Results: Light microscopic examination of group II showed the degenerative changes that caused by MSG such as dark and shrunken Purkinje and granule cells. This was confirmed by electron microscope which revealed bizarre Purkinje nucleus surrounded by perineural space and abnormal dark granule cells surrounded by myelinated nerve fibers with detached myelin sheaths. Statistical analysis of Purkinje cell number between the two groups showed significant decrease in group II compared to group I.


Conclusion:MSG has a degenerative effect on rats' cerebellum, and it can trigger neurotoxicity.
