Cerebroprotective Activity Of Hispolon And Its Derivative In Cerebral Ischemic Reperfusion Injured Rats

Abstract

Aim Of This Study Is To Determine The Cerebroprotective Effect Of Hispolon And Hispolon Mono Methyl Ether (Hmme) Using I/R Induced Ischemia In Rats. Animals Were Divided To Control, I/R Control, Hispolon 10mg/Kg And 20mg/Kg, Hmme 10mg/Kg And 20mg/Kg Bd.Wt. P.O. Treatment Was Given To The Animals For A Period Of 14 Days. On 15th Day Transient Global Cerebral Ischemia Was Induced By 30-Minute Occlusion Of The Bilateral Common Carotid Artery, Followed By 4-Hour Reperfusion, At The End Of The Experiment Infarct Size Followed By Evaluation Of Morphological, Biochemical And Histopathological Evolutions. The Elevated Serum Biochemical Parameter Teac And Frap And Decreased Tbrs Were Observed With Hispolon And Hmme. The Oxidative Stress Markers Such As Sod, Cat, Gpx And Gsh Were Found Be Enhanced And Reduced Levels Of Mda, Ldh, Protein Carbonyl And Nitrites Were Observed With Hispolon And Hmme Treatment In I/R Induced Ischemic Rats. Inflammatory Mediators Tnf Α And Il 6 Were Reduced And Anti Inflammatory Mediator Il 10 Was Found To Enhance In I/R Induced Rats Treated With Hispolon And Hmme. Treatment With Hispolon And Hmme Showed Decreased Brain Water Content And Infarct Volume And Ameliorated The Damaged Structure And Necrosis Of Hippocampus And Cerebral Cortex.
