Peroxidase is an isoenzyme marker for Prarylenchus zeae resistance in sugarcane clones

Abstract
Globally, the migratory endoparasitic nematode Pratylenchus is a primary nematode pathogen of sugarcane fields. The breeding variety for nematode resistant is the most promising and cost-effective methods when compared with all other nematode management practices. The present studies were aimed to detect defense responsive mechanisms involved in sugarcane - P. zeae interaction. The native PAGE analysis of peroxidase activity was carried out in P.zeae infected 11 susceptible and 11 resistant sugarcane clones. The peroxidase isoform of Rf value of 0.36 found in resistant clones notwithstanding in susceptible clone. Therefore, the specific peroxidase isoform of sugarcane clones has the potential to be used as a resistant marker to P. zeae. The peroxidase loci (0.36) segregation pattern is analyzed by conventional breeding sugar cane progenies of resistance x resistance and resistance x susceptible crosses. The progenies are found lesser number of resistant markers and conferred as a recessive gene(s) and was later confirmed through examination of selfed progeny of resistant clone Co 6304.
